mTOR is the target molecule of rapamycin (trade name: Sirolimus) in mammals and it has been recognized as a central molecule in the signaling pathway regulating proliferation and cell growth. Since DNA replication is also essential for proliferation, regulation of cell growth and amino-acid perception it is also centrally related to mTOR and deeply involves in the biosynthesis of nucleic acids. CAD (carbamoyl phosphate synthetase, aspartate transcarbamoylase, dihydroorotase) was discovered as a result of a survey for molecules that are related to nucleic acid biosynthesis in a molecular complex with raptor and mLST8. CAD has an important role in the pathway of pyrimidine biosynthesis. CAD is an enzyme that catalyzes the pyrimidine biosynthesis and itself is regulated by phosphorylation of other protein kinases. mLST8 bridges between CAD and mTOR and CAD plays a role as a signaling molecule in the mTOR pathway by a cross-interaction with mLST8.
