Although lipids are not soluble in water, apolipoproteins are carrier proteins binding to lipids forming lipoprotein particles, that are water-soluble and can be carried through water-based circulation (i.e., blood, lymph). Apolipoproteins are classified according to their forms and functions, and there are six major classes and several sub-classes. ApoB (ApolipoproteinB) is the primary apolipoprotein of Low-Density-Lipoproteins (LDL) and is considered to reflect changes in LDL levels in blood. There are two types of apolipoproteins B. The first is ApoB-100, which correlates with cholesterol levels and is mainly released as part of LDL and VLDL. The second is ApoB-48, which is part of chylomicrons and correlates with tryglyceride levels. In general, concentration of ApoB in blood is determined as sum of ApoB-100 and ApoB-48.
This kit measures solely ApoB-100 in plasma samples by using ApoB-100 specific antibodies, that don't cross react to ApoB-48. This highly sensitive sandwich ELISA can also measure ApoB-100 in cell culture media or various fractionated lipoprotein samples.
