Intended Use

This for Research Use Only product is intended to assist in the detection of previous ethanol exposure by the quantitative determination of different phosphatidylethanol (PEth) species in whole blood, using LC-MS. The product is not intended for self-testing, companion diagnostic use, or as a fully automated test.

Background

Phosphatidylethanols (PEths) are abnormal phospholipids formed only in the presence of ethanol, via phospholipase D-mediated transphosphatidylation of phosphatidylcholine in red blood cell membranes [1,2]. Among over 40 known PEth homologues, POPEth (PEth 16:0/18:1) and PLPEth (PEth 16:0/18:2) are the most abundant and analytically robust [3]. They can be detected in whole blood for up to approximately 2–4 weeks after the last alcohol intake, with concentrations correlating strongly with the amount and frequency of consumption [4–5]. Quantification by LC–ESI–MS/MS offers high analytical specificity and sensitivity, with sub-ng/mL detection limits.

LC-MS Method

PLPEth and POPEth are determined from human whole blood by UHPLC with negative ion electrospray LC-MS/MS. Prior to the LC-MS/MS analysis, a sample clean-up is performed to remove the sample matrix and to spike with the internal standard.
